台灣國產柳橙皮甲醇萃取物之成分分析與抗氧化性研究

Study on Analysis of Components and Antioxidative Activity in Orange Peel Extract Cultivated in Taiwan

摘要


本試驗主要為利用甲醇萃取方式,萃取台灣國產柳橙果皮,評估萃取液之總多酚、總類黃酮含量與抗氧化特性,並以HPLC分析其類黃酮組成,以期可以開發成天然之抗氧化劑。結果顯示柳橙果皮萃取液之總多酚及總類黃酮含量分別為53.35mg/mL、5.47mg/mL。而類黃酮組成方面,柳橙果皮萃取液之橘皮苷(hesperidin)含量最多(689.31μg/mL),其次為柚皮苷(naringin)(134.62μg/mL)與槲皮酮(quercetin)(33.52μg/mL),山茶酚(kaempferol)為20.11μg/mL。在抗氧化方面,柳橙果皮萃取液具有良好之DPPH自由基清除力、還原力與亞鐵離子螯合能力,且隨著萃取液濃度之增加其能力亦有增加之趨勢,硫鐵法測定同樣顯示出柳橙果皮萃取液具有抑制亞麻仁油酸過氧化之效果。本研究之結果說明柳橙果皮萃取物具有開發成天然抗氧化劑之潛力。

In a current study, Taiwan cultivating orange peel was extracted using methanol and evaluated its contents of total phenolic compounds and total flavonoids, and antioxidative activity. Also quantitation of flavonoids compounds were carried out by HPLC in order to develop a natural antioxidant. The results showed that 53.35 mg/mL of total phenolic compounds and 5.47 mg/mL of total flavonoids were observed in orange peel extracts. In flavonoids compositions, the highest content of hesperidin (479.62μg/mL) was found, and the second were naringin (114.53μg/mL) and quercetin (46.02μg/mL). Kaempferol (20.11μg/mL) was also determined in orange peel extracts. In antioxidative properties, orange peel extracts exhibited strong scavenging activity of DPPH free radical residue, reducing capacity and Fe(superscript 2+) chelating ability, which contributed to inhibitory activity of linoleic acid peroxidation. Dose-depending activity was also discovered in antioxidative properties. The present results suggest that orange peel extract possess high potential to develop as natural antioxidant.
