Moringa Oleifera Leaf Ameliorates Fat Accumulation in Rats Fed High-Sucrose Water

Purpose: This study investigated the effects of Moringa oleifera leaf (MOL) on fat accumulation in rats fed high-sucrose water. Methods: Three-week- old male Wistar rats were randomly divided into two groups: a control group (n=11) and a group fed MOL powder (6% in the test diet) (n=11). All had free access to the test diet and to 30% sucrose water. After 8 weeks of feeding, the rats were sacrificed by carbon dioxide asphyxiation after an overnight fast. Blood and all tissues were collected and weighed for further analysis. Results: Liver, retroperitoneal fat and epididymal fat pad weights had decreased significantly with MOL feed. Rats in the MOL group also had significantly decreased serum and liver total cholesterol (TC) levels and hepatic triglyceride (TG) levels, and a marked increase in fecal cholesterol excretion as compared to the control group. MOL supplementation significantly reduced liver Xreceptor-α (LXR-α), carbohydrate response element-binding protein (ChREBP), peroxisome proliferator-activated receptor gamma (PPAR-γ) and sterol regulatory element-binding protein- 1c (SREBP-1c) messenger RNA (mRNA) levels; the reductions were approximately 48%, 37%, 44%, and 47%, respectively. Conclusion: MOL is an edible vegetable; rats fed MOL had significant reductions in body weight and fat accumulation. Supplementation with MOL ameliorated sucrose-induced hypercholesterolemia; this may be related to the increase in fecal cholesterol excretion.

目的:探討添加辣木葉(Moringa oleifera leaf, MOL)是否具有緩和高蔗糖水誘導大鼠脂肪堆積效應。方法:將三週齡Wistar品系公鼠依體重隨機分成兩組,包括糖水控制組(30%蔗糖水)與辣木葉補充組(30%蔗糖水+飼料含6%辣木葉粉),每組11隻,30%蔗糖水、水與飼料均令其自由食用,飼養8週後,禁食後以CO2窒息犧牲,取血液與組織進行相關分析。結果:辣木葉餵食顯著降低大鼠肝臟、腹膜後與副睪脂肪量,顯著降低血清、肝臟之膽固醇含量與肝臟三酸甘油酯(Triglyceride, TG)濃度,增加糞便膽固醇排出,肝臟轉錄因子之mRNA方面,補充辣木葉明顯降低liver X receptor(LXR)、Carbohydrate response element-binding protein (ChREBP)、peroxisome proliferator-activated receptor gamma(PPAR-γ)與sterol regulatory element-binding protein-1c(SREBP-1c)mRNA量,分別為糖水組的48%、37%、44%與47%。結論:辣木葉是種可食用的蔬菜,辣木葉餵食大鼠之體重與脂肪塊顯著降低,補充辣木葉可緩和高蔗糖水誘導大鼠高膽固醇血症,可能與增加糞便膽固醇排出有關。
