香瓜茄水萃物於糖化終產物刺激下對於RIN-m5F cells之影響

摘要


香瓜茄為澎湖地區特有的茄科多年生草本植物,本實驗室先前已發現香瓜茄對於糖尿病小鼠具有延緩其併發症病程之作用。在台灣地區糖尿病是影響重大的慢性疾病之一,細胞長期處於高血糖環境下易有氧化損傷,且長期的高血糖會造成糖化終產物(Advanced glycation end products;AGEs)的生成,氧化壓力增加進而誘發發炎反應,造成β細胞功能失調、數目減少、萎縮及死亡等情形。故本研究以細胞模式探討在糖化終產物的環境下對於大鼠胰島細胞(RIN-m5F cells)的影響,並觀察香瓜茄水萃物是否具有保護的作用。實驗將RIN-m5F cells分組為控制組(Control)、高糖組(225mM glucose)、AGEs處理組、AGEs+香瓜茄處理組(不同劑量香瓜茄水萃物)培養48小時後收集細胞樣本進行分析。結果顯示在AGEs的刺激下添加香瓜茄水萃物可幫助細胞恢復胰島素的分泌能力、增加細胞之存活率、降低氧化壓力、減少細胞發炎、凋亡和降低AGEs其接受器(RAGE)之mRNA表現量。因此根據結果推論RIN-m5F cells在AGEs的刺激下,添加香瓜茄水萃物可以降低氧化壓力的傷害、減少細胞發炎、凋亡,保護細胞因AGEs的添加而產生的傷害。

Solanum muricatum Ait (pepino) is a popular plant fruit in Penghu island, Taiwan. Our previous study reported that pepino intake could attenuate diabetic complication in type 2 diabetic BALB/c mice. Advanced glycation end products (AGEs) play an important role in the development and progression of diabetes-related complications. AGEs could induce β-cell dysfunction, and promote diabetic deterioration. The present study was designed to investigate the effects of an aqueous extract from pepino (AEP) against AGEs induced RIN-m5F cells dysfunction. RIN-m5F cells was cultured under control, high glucose (225mM glucose), AGEs, AGEs plus AEP (different dose of AEP) for 48 hours. Results showed that AEP treatments recovered the ability to secret insulin of RIN-m5F cells, increased cell viability, decreased oxidative stress, reduced inflammation, and reduced mRNA expression of AGEs receptor (RAGE). These data suggested that pepino was able to exert protective activities against AGEs induced injury in RIN-m5F cells dysfunction by decreasing oxidative stress, reduced inflammation, and inhibited cell apoptosis.
