摘要


甲狀腺屬於內分泌組織,其所分泌的甲狀腺素(T4、T3)可調節新陳代謝以及生長與發育等功能。目前已知,藉由喪失調控細胞週期的進行以及細胞凋亡的作用造成細胞不正常生長是形成甲狀腺腫瘤的主要原因。本研究利用組織免疫染色來偵測與細胞週期有關的蛋白如:p27及skp2或是與細胞凋亡有關的蛋白如:XIAP及Survivin分別在甲狀腺乳突癌(Papillary thyroid carcinoma)、甲狀腺濾泡癌(Follicular thyroid carcinoma)、結節性甲狀腺腫瘤(Nodular goiter)及濾泡性腺腫瘤(Follicular adenoma),等四種腫瘤細胞上的表現情形。結果顯示p27蛋白在各種腫瘤組織中表現極少數,相對而言skp2蛋白於不同的甲狀腺腫瘤組織皆有大量表現。XIAP及Survivin主要表現於甲狀腺乳突癌及甲狀腺濾泡癌而在腺腫瘤組織則幾乎沒有表現。本實驗顯示,未來可利用XIAP及Survivin來當作區別惡性或是良性甲狀腺腫瘤的依據之一。

The thyroid gland, one endocrine system, can produce thyroid hormone (T4 and T3) which regulates many cellular functions such as metabolism, cell growth and development. To date, deregulation of cell cycle progression and apoptosis rendered cell to be abnormal growth is the major event resulting in thyroid cancer formation. In this study, we have performed immunohistochemical assay to detect the expression pattern of two cell cycle regulators: p27 and skp2 and two apoptosis proteins: XIAP and survivin in papillary thyroid carcinomas, follicular thyroid carcinomas, follicular adenoma, and nodular goiter. We found that p27 was rarely expressed in all samples. On the other hand, skp2 was highly expressed in almost, if not all, samples. XIAP and survivin was expressed higher in carcinomas than in adenomas. Taken together, our result suggested that XIAP and survivin may serve a marker for distinguishing carcinoma from adenoma.
