傳統中藥龜鹿二仙膠對化療小鼠的運動表現、 心肌保護與骨鬆緩解作用

Effects of the Traditional Chinese Medicine, Guilu Erxian Gum, on Exercise Performance, Myocardial Protection and Osteoporosis Relief in Chemotherapy Mice

摘要


目的:探討增補龜鹿二仙膠對小鼠注射癌症化療藥物艾黴素後運動表現、心肌保護與降緩骨質疏鬆的效果。方法:將21隻ICR小鼠分為控制組(control, CON, N=4)、中藥增補組(control+Guilu Erxian Gum, CON+GEG, N=5)、化療處理組(doxorubicin, DOX, N=6)及中藥+化療組(doxorubicin+Guilu Erxian Gum, DOX+GEG, N=6)。CON+GEG和DOX+GEG兩組在第二、三、四和七週時早晚各管餵100毫克/公斤/次劑量的龜鹿二仙膠,而DOX和DOX+GEG兩組則於第二、三和四週時每週腹腔注射兩次3毫克/公斤/次劑量的艾黴素。四組皆在第七週進行游泳力竭測驗,第八週犧牲後採集血液、尿液、股骨和心肌組織進行後續分析。所得資料以獨立樣本單因子變異數分析檢驗四組游泳力竭時間、血流通量、骨量百分比及第八週時體重的差異。結果:CON和CON+GEG兩組游泳力竭時間顯著高於DOX和DOX+GEG組;CON+GEG組血流通量和抗氧化能力顯著高於CON組;DOX組骨質密度流失且心肌組織表現出較大氧化壓力與發炎反應,而DOX+GEG組具有緩解效果。結論:龜鹿二仙膠可以提升血液循環、心肌組織抗氧化能力,以及緩解化療對體重、運動表現、發炎反應和骨質密度的負面影響。

Purpose: To investigate the effects of Guilu Erxian Gum on exercise performance, myocardial protection and osteoporosis relief in chemotherapy mice. Methods: 21 ICR mice were assigned to control group (CON, N=4), Guilu Erxian Gum supplement group (CON+GEG, N=5), doxorubicin chemotherapy treatment group (DOX, N=6), and Guilu Erxian Gum and doxorubicin group (DOX+GEG, N=6). CON+GEG and DOX+GEG group were received 100 mg/kg/time of Guilu Erxian Gum twice a day in the 2nd-4th and 7th weeks. DOX and DOX+GEG group were injected 3 mg/kg/time of doxorubicin twice a week in the 2nd-4th weeks. All groups performed exhausted swimming test in the 7th week. In the 8th week, mice were sacrificed, and their blood, urine, femurs, and myocardial tissue were collected for analysis. Independent one-way ANOVA was used to analyze the differences among groups on exhausted swimming time, blood flow, bone density and body weight of 8th week, respectively. Results: The swimming exhaustion time of CON and CON+GEG groups were significantly higher than that of DOX and DOX+GEG groups. The blood flow and antioxidant capacity of CON+GEG group were significantly higher than those of CON group. The bone density of DOX group was lost and its myocardial tissue showed oxidative stress and inflammation, however, DOX+GEG group had an alleviated effect. Conclusion: GEG can improve blood circulation, antioxidant capacity of myocardial tissue, and alleviate the side effects of chemotherapy on body weight, exercise performance, inflammation, and bone density.
