摘要


輸血早已成為醫療行為的重要一環,不但可以成為疾病預防、診斷的重要因素,也可以利用血液進行治療治療,近年來免疫醫學的突破發展,人類可有效使用白血球來進行疾病診斷與治療。過去的文獻中證實白血球會引發一些併發症使身體不適,因此分離全血中的白血球是必要的,並且在輸血過程中移除白血球可以進行疾病預防並降低副作用。而未來回收白血球可以進行再生醫學領域的發展,其重要性逐漸受到重視。本研究目的為設計與製備溫感型聚丙烯纖維薄膜於人體白血球分離及回收。實驗規劃製備出具溫感特性之共聚高分子,並將其改質於纖維膜表面,使聚丙烯纖維膜改質後更加親水,且提高材料生物相容性,以研究其對於白血球捕捉與釋放的控制,並探討改質後的膜材料對於白血球捕捉與釋放的效能。本研究結果顯示,改質後的聚丙烯纖維膜過濾3mL白血球濃厚液(WBC),證明此膜材系統可提供極高的白血球捕捉率,並且有效的回收白血球。此研究工作證明該共聚高分子所製備與改質的聚丙烯纖維薄膜系統,可以實現人體血液中的白血球分離純化及回收,未來也具有潛力來發展疾病診斷與細胞免疫療的應用。

Blood transfusion has long been an important part of medical practice. It can not only become an important factor in disease prevention and diagnosis, but also can be used for treatment and treatment. In recent years, the breakthrough development of immune medicine has enabled humans to effectively use white blood cells for disease diagnosis and treatment. It has been demonstrated in the literature in the past that leukocytes can cause some complications and cause discomfort, so separation of leukocytes from whole blood is necessary, and the removal of leukocytes during transfusion can prevent disease and reduce side effects. In the future, the recovery of white blood cells can be used for the development of regenerative medicine, and its importance has gradually been paid attention to. The purpose of this study was to design and prepare a temperature-sensitive polypropylene fiber film for separation and recovery of human leukocytes. The experimental plan prepares a copolymer polymer with temperature-sensing properties and modifies it on the surface of the fiber membrane to make the modified polypropylene fiber membrane more hydrophilic and improve the biocompatibility of the material to study its effect on the capture and release of leukocytes. control, and explore the efficiency of the modified membrane material for leukocyte capture and release. The results of this study show that the modified polypropylene fiber membrane filters 3 mL of white blood cell concentrate (WBC), which proves that this membrane system can provide a very high white blood cell capture rate and effectively recover white blood cells. This research work proves that the polypropylene fiber membrane system prepared and modified by the copolymer can realize the separation, purification and recovery of white blood cells in human blood, and also has the potential to develop applications in disease diagnosis and cellular immunotherapy in the future.
