本研究建立培養樹豆三種部位(子葉、胚軸、根部)癒合組織,以總酚含量、總黃酮含量及DPPH自由基清除能力三種方式分析其抗氧化能力,並與成熟的樹豆及黃豆種子比較;再利用genistein和daidzein兩種標準品,檢測這五種檢品中大豆異黃酮的含量。結果顯示:1、樹豆癒合組織的生長倍率,以根部癒合組織培養14天成長13.8 ± 1.11倍為最快速。2、比較五種檢品的乙醇萃取物,發現樹豆根部癒合組織總酚含量(7.45 ± 0.14 mg gallic acid eq./g D.W.)、DPPH自由基清除力在稀釋比率為1/4時,具有接近90 %的清除能力,其抗氧化力為五種檢品中表現最佳者。3、使用daidzen和genistein為標準品檢測五種檢品酸解後異黃酮含量,發現三種不同部位的樹豆癒合組織中都只有檢測出genistein,樹豆種子則是都未檢出;在總異黃酮含量的部份以樹豆子葉癒合組織最好,是黃豆種子的6倍。
Three kinds of explants(cotyledon, hypocotyl and root) from Cajanus cajan were used to established callus culture. Ethanolic extract prepared from five plant materials, callus from cotyledon, callus from hypocotyl, callus from root, Cajanus cajan seeds and Glycine max seeds were examined for their phenolic contents, flavonoids contents and DPPH radical - scavenging activity. The content of isoflavone (genistein and daidzein) in the ethanolic extract was determined by high-performance liquid chromatography. The multiplication of callus was very fast and the biomass increase of callus from root reached to 13.8 ± 1.11 folds within 14 days culture. Among five plant materials, the ethanolic extract of root originated callus expressed high total phenolic contents (as gallic acid equivalents: 7.45 ± 0.14 mg /g D.W.) and high DPPH radical- scavenging activity (90 % at quarter strength of extract) . The content of isoflavone of callus only expressed genistein and the callus from cotyledon showed the highest total isoflavone contents which was 6 times of Glycine max seeds.
