利用細胞膜熱穩定性技術篩選高耐熱性葉用蘿蔔

Selection High Heat Tolerance Leafy Radish Cultivars/Lines by Cell Membrane Thermostability

摘要


葉用蘿蔔(Raphanus sativus L. var. oleiformis Pers.)耐熱育種為產業上所需求,篩選出具有耐熱潛力之基因型則為育種的第一步。本試驗利用電解質滲漏技術測量葉用蘿蔔葉片和子葉細胞膜熱穩定(CMT),並觀察其植株生育表現。結果顯示不同品種(系)之葉用蘿蔔葉片和子葉之熱傷害值與水浴處理溫度之間呈現一S曲線關係,曲線反折點約在水浴溫度46~50℃。50℃處理之熱傷害值接近曲線反折點,且對於品種(系)間耐熱性檢測有較佳的敏感度。選六品種(系)做進一步植株生育表現之觀察,結果顯示,六品種(系)之植株乾重、淨光作用、葉綠素螢光值(Fv/Fm)和葉綠素含量在35℃高溫下減少,反之蒸散速率、過氧化氫含量及丙二醛含量則增加,且呈現品種(系)間的差異。迴歸分析結果則指出植株乾重與葉綠素螢光值因35℃所減低之比例和50℃水浴之熱傷值呈現二次相關性。

Breeding for heat-tolerance of leafy radish (Raphanus sativus L. var. oleiformis Pers.) is of commercial interests, and the identification of potential genotypes is the first step to onset the program. An electrolyte leakage technique was applied to measure cell membrane thermostability (CMT) of leaf and cotyledon discs of leafy radish cultivars/lines. A sigmoidal curve relationship existed between the relative injury (RI) value in leaf and cotyledon tissue discs of each cultivar and water bath temperatures. The RI values near the midpoint of the sigmoid response curve fall within 46~50℃. A single treated temperature 50℃ gave RI value near the inflection point and showed good sensitivity in detecting the genotypic difference in heat tolerance. Six cultivars/lines with different RI value were chosen for further observation on plant growth performance. Plant dry weight, net photosynthesis, chlorophyll content and chlorophyll fluorescence value (Fv/Fm) were decreased while transpiration rate, H2O2 and malondiadehyde content were increased at 35℃ in comparison to 25℃. Plant dry weight and Fv/Fm reduction ratio at 35℃ versus their associated RI value at 50℃ showed a quadratic relationship.
