香瓜茄水萃物於糖化終產物刺激下 對於內皮細胞之影響

The effect of an aqueous extract from Solanum muricatum Ait on endothelial cells after advanced glycation end products (AGEs) treatment

摘要


糖尿病已是影響台灣地區最重要的慢性疾病之ㄧ,長期的高血糖會加速糖化終產物的產生,進而影響內皮細胞,造成內皮細胞功能失調而發生動脈粥狀硬化等血管病變。香瓜茄屬於茄科的多年生草本植物,在台灣有零星栽培,其中以離島的澎湖最多,本實驗室先前已發現香瓜茄對於糖尿病小鼠具有延緩併發症病程之作用,故本研究以細胞模式探討在糖化終產物(Advanced Glycation End Products ; AGEs)刺激下對於人類臍靜脈內皮細胞(Human Umbilical Vein Endothelial Cells ; HUVECs)的影響,並觀察香瓜茄水萃物是否具有保護的作用。本實驗將HUVECs分別培養於控制組(control)、AGEs處理組(2.35mg/ml AGEs)、AGEs+香瓜茄處理組(2.35mg/ml AGEs+不同劑量香瓜茄水萃物)培養48小時後收集細胞樣本進行分析。結果顯示在AGEs的刺激下添加香瓜茄水萃物可增加細胞之存活率、減少細胞凋亡、降低氧化壓力、下降血管細胞黏著分子(Vascular cell Adhesion Molecule-1; VCAM-1)和細胞間黏著分子(Intercellular Adhesion Molecule-1; ICAM-1) mRNA 的表現量。因此根據結果推論HUVECs在AGEs刺激下,添加香瓜茄水萃物可減少細胞死亡、降低氧化壓力的傷害,下降內皮細胞黏附因子的表現以減少發炎細胞的附著,進而延緩糖尿病相關之血管病變與併發症的發生。

Advanced glycation end products (AGEs) have been recognized as one of the most important factors responsible for the progression of diabetes. AGEs could cause harmful effects upon endothelial cells, which leads to vascular dysfunctions. Solanum muricatum Ait (pepino) is a popular fruit in Penghu island, Taiwan. Our previous study indicated that pepino intake was able to attenuate diabetic complications in type 2 diabetic BALB/c mice. The present study was designed to investigate the protective effect of an aqueous extract from pepino (AEP) against AGEs induced endothelial dysfunction in human umbilical vein endothelial cells (HUVECs). HUVECs were cultured under normal, BSA(2.35mg/ml BSA), AGEs (2.35mg/ml AGEs), AGEs plus AEP10 (2.35mg/ml AGEs+10μg/ml AEP), AGEs plus AEP20 (2.35mg/ml AGEs+20μg/ml AEP) for 48 hours. Results showed that AEP treatments exhibited anti-apoptotic effects, increased cell viability, decreased oxidative stress, reduced ICAM-1 and VCAM-1 mRNA expression. These data suggested that pepino could possess protective activity on AGEs-induced endothelial dysfunctions, including anti-apoptosis, anti-inflammation and anti-oxidative stress .Pepino might be considered as an alternative functional food for the prevention and therapy to alleviate diabetic vascular complications.
