國產大豆抗氧化活性之研究

Antioxidant Activity of Taiwan Soybeans

摘要


本研究選擇7個臺灣目前推廣之黑豆及毛豆為材料,針對其抗氧化活性進行研究。黑豆及毛豆萃取物的抗氧化活性分析結果,黑豆臺南三號萃取物(4 mg mL^(-1))的抗脂質氧化能力與人工合成之抗氧化劑BHT(1 mg mL^(-1))相當。清除2,2-diphenyl-1-picrylhydrazyl (DPPH)自由基的能力,也是以黑豆臺南三號優於其他供試品種。各黑豆及毛豆萃取物的總酚含量,以黑豆臺南三號為首(4-9 mg 100 g^(-1))、臺南一號及臺南五號次之(6 mg 100 g^(-1))。令人意外的發現,黑豆高雄七號的總酚含量是全部測試品種之中最低者。利用普魯士藍(Prussian blue)之生成量為還原力測定指標,黑豆臺南三號萃取物的還原力居首位,大約為BHT (1 mg mL^(-1))的70-80%。毛豆高雄五號的螯合亞鐵離子能力最好,黑豆臺南三號最差。綜合以上的試驗結果,國產黑豆與毛豆具有相當的抗氧化活性,其中黑豆臺南三號的抗脂質氧化與清除DPPH自由基的能力均優於其他豆子,唯螯合亞鐵離子的能力不佳。

Seven Taiwan soybean cultivars were screened for antioxidant activity. The antioxidant activity against lipid peroxidation of soybean organic extracts (4 mg mL^(-1)) were measured using the ferric thiocyanate (FTC) method with synthetic antioxidant BHT (1 mg mL^(-1)) as control. TN3 extract showed the best and equivalent antioxidant activity against lipid peroxidation to BHT. The free radical scavenging activity was determined by the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical-scavenging method. The activity exhibited in the TN3 extract was significantly higher than that of other extracts. The total phenolic content (TPC) was determined according to the Folin-Ciocalteu method. TN3 was found to contain the highest phenolic content (4-9 mg 100 g^(-1)) and followed by TN1 and TN5 (6 mg 100 g^(-1)). Surprisingly, the lowest amount of phenolic was found in the black seed coat cultivar of KS7 extract. The reducing power of the TN3 extract measured by the formation of Prussian blue displayed activity at 70 to 80% of BHT. Additionally, the comparative ability of soybean extracts to chelate iron was determined by the decolorization of a FeCl2/Ferrozine solution. The ability of iron chelation by the KS5 was significantly higher than that of other extracts. TN3 extract showed the lowest ability of iron chelation. This study provides further evidence that the organic extract of TN3 is an effective antioxidant and scavenger of radical but much less effective in iron chelation.
